Key component in mother's egg critical for survivial of newly formed embryo

ScienceDaily (Mar. 29, 2012) ? An international team led by scientists at A*STAR's Institute of Medical Biology (IMB) discovered that a protein, called TRIM28, normally present in the mother's egg, is essential right after fertilisation[1], to preserve certain chemical modifications or 'epigenetic marks' on a specific set of genes. This newly published study paves the way for more research to explore the role that epigenetics might play in infertility.

Previous studies have shown that both nuclear reprogramming as well as 'imprinting' are vital for the survival and later development of the embryo. However, the underlying mechanisms governing the intricate interplay of these two processes during the early embryonic phase have not been clear, until now.

Nuclear Reprogramming

Immediately after fertilisation, the majority of the 'epigenetic marks' on the DNA from the sperm and egg cells are erased. The erasure process, termed nuclear reprogramming, allows the genes from the parents to be reset so that the early embryonic cells can develop into any cell types of the body.

Genomic Imprinting

On the other hand, certain 'epigenetic marks' on a particular set of genes, some from the mother and some from the father must be preserved. These genes are said to be "imprinted" by their parent of origin and preservation of these marks is critical for the survival of the newly formed embryo. Expression of these imprinted genes at the appropriate levels ensures proper development of the embryo. If the 'epigenetic marks' on the 'imprinted' genes are not protected, severe and multiple developmental defects occur in the embryo.

Using genetically identical mice from an inbred mouse strain[2], Drs. Davor Solter and Barbara Knowles, Senior Principal Investigators at IMB, observed that none of the embryos resulting from the fertilisation of eggs lacking TRIM28 survived. The embryos died at varying stages of development, and had very different developmental defects. Given that most genetic diseases attributed to the lack of a single gene have consistently similar defects in every affected individual, the highly varied abnormalities observed in the maternal TRIM28-deficient, yet genetically identical mice, were peculiar.

To further elucidate these findings and the role of TRIM28 during nuclear reprogramming, Dr. Daniel Messerschmidt, the first author of this paper, collaborated with Dr. Anne Ferguson-Smith, a visiting Professor at A*STAR's Singapore Institute for Clinical Sciences (SICS), from the University of Cambridge, UK. Together, they conducted a comprehensive study on the epigenetic state and the level of expression of a group of imprinted genes known to be important for development. Using highly sophisticated microarray analysis and advanced biochemical techniques, they found that not only is the presence of TRIM28 protein important during this critical period, but the timing and amount of TRIM28 made available to the newly formed embryo greatly impacts the expression of these imprinted genes at later developmental stages.

Genomic imprinting was first described by Dr. Davor Solter at the Wistar Insitute and by Dr. Azim Surani at Cambridge University three decades ago. Recently, Dr Solter suggested that the interplay between nuclear reprogramming and genetic imprints could be explained by a simple analogy from school where the teacher writes a long string of formulas on the black board. At the end of the class he marks stretches to be preserved for the next day's lecture (these are the imprinted marks) and asks the cleaning lady to erase the rest (she performs the reprogramming).

Dr Solter said, "For the longest time, we didn't know why some bits of the formulas escaped erasure. Through this study, the mystery is finally revealed! We now know TRIM28 instructs the 'cleaning lady' which part of the formulas she should preserve."

Dr. Azim Surani, who was not involved in this study, said "This elegant study on TRIM28 provides significant mechanistic insights on how maternal factors in eggs ensure faithful epigenetic inheritance that plays a critical role during development. These findings are relevant not only for the widely practised in vitro fertilisation of eggs for the treatment of human infertility, but also more generally for highlighting the importance of epigenetic mechanisms in development and disease."

Dr. Barbara Knowles, the senior author of this paper said, "Lack of TRIM28 in their eggs could explain why some women consistently suffer from multiple failed pregnancies where embryos die at different time points, manifesting multiple, different abnormalities."

Prof Birgitte Lane, Executive Director of IMB, said, "Understanding the role of differential epigenetic modifications is important for the study of human diseases and development. This study has unravelled a critical component essential for nuclear reprogramming. The mechanistic insights revealed have far-reaching clinical implications for treatment of diseases with patient-specific cell therapies. This finding demonstrates how discoveries of fundamental research can shed light on biological questions to open new avenues of formulating therapeutic strategies for the benefits of patients."

A University of Adelaide study has revealed that inducing labor in pregnant women when it's not medically necessary is more likely to result in complications at birth.

Elective induction is becoming more common around the world, with many women being induced for social and other non-medical reasons.

Dr Rosalie Grivell from the University of Adelaide's Robinson Institute has studied the data of more than 28,000 births from across South Australia, from 2006 to 2007.

She compared cases in which women had undergone spontaneous onset of labor, induction of labor for recognized medical reasons, and induction of labor for "non-recognized" reasons.

Compared with women who entered labor spontaneously, induction for non-recognized reasons was associated with a 67% increased chance of requiring a cesarean section.

It also significantly increased the chance of the newborn infant requiring nursery care in a Special Care Baby Unit (an increased risk of 64%) or requiring treatment (an increased risk of 44%) compared with infants born following spontaneous onset of labor.

"Our research is aimed at better understanding the optimal timing and management of labor and birth for women with an uncomplicated pregnancy," Dr Grivell said.

"We hope our findings will increase awareness of the potential harmful effects that elective induction can have on both women and their infants. In the absence of serious maternal or fetal problems or a medical recommendation, induction of labor is best avoided."

Dr Grivell said the lowest risk of adverse complications both for mother and baby occurred with the spontaneous onset of labor between 38 and 39 weeks.

"While a natural birth is not always possible for women who already have complications in pregnancy, the results of this study suggest that for women whose pregnancy is uncomplicated, awaiting the spontaneous onset of labor is best," Dr Grivell said.

WASHINGTON (AP) ? Republican Sen. Marco Rubio of Florida endorsed Mitt Romney as the party's presidential nominee Wednesday night, saying it's increasingly clear that Romney will win the nomination and that continuing the primary fight will only damage the effort to defeat President Barack Obama.

Rubio, a freshman senator and tea party favorite, said he's convinced that Romney will govern as a conservative and will be superior to Obama as a president.

"The quicker we can get this campaign on that focus ? focused on the president's record, on the alternative that we offer ? the better off we're going to be as a movement but also the better off the country's going to be," Rubio said during an interview with Fox News.

Rubio had pledged to stay neutral in the race. On Wednesday he called a floor fight over the nomination at the party convention in August "a recipe to deliver four more years to Barack Obama."

Rubio's endorsement followed that of former Florida Gov. Jeff Bush and came a day before former President George H.W. Bush was to back Romney, another sign that more and more party leaders are falling in line behind Romney rather than support rivals Rick Santorum, Newt Gingrich or Ron Paul. Gingrich has scaled back his campaign, saying he wants to focus on winning the nomination at the convention if Romney fails to win enough delegates during the primary race.

Rubio has been mentioned as a potential running mate on the GOP ticket. He told Fox News that his heart, mind and focus remain on the Senate and that he doubts he would be asked to join a Romney ticket.

Fan declared brain dead after violence in Brazil

SAO PAULO (AP) -A Palmeiras fan was declared brain dead on Tuesday, the second casualty from a confrontation involving nearly 500 people from rival groups last weekend.

The Sao Camilo hospital said the 19-year-old fan remains on a ventilator but will not recover after receiving head injuries in the fighting on Sunday. His name was not immediately released by the hospital.

On Sunday, 21-year-old Palmeiras supporter Andre Alves died after being shot in the head in the confrontation. Two other fans remain hospitalized, a 17-year-old with head injuries and a 23-year-old who was shot in the hip and needed surgery.

The announcement that the fan was brain dead came as authorities decided to close the headquarters of the rival fan groups involved in the fighting - Palmeiras' Mancha Verde and Corinthians' Gavioes da Fiel. The Sao Paulo state football federation indefinitely banned them on Monday from entering stadiums.

Authorities seized computers and other material that could bring more information about those involved in the fight, and detained several members suspected of participating in the confrontation. Iron bars and other possible weapons used in the brawl were also seized.

Police said one of Alves' brother, a vice president at the Mancha Verde, was shot in the leg during another fight last year.

The confrontation on Sunday raised concerns about escalating fan violence in Brazil, and authorities said they will have to take action to keep the fighting from spreading with the country staging the 2014 World Cup. Corinthians' stadium will host the World Cup opener in 2014.

"We are against this type of violence because everybody loses," said former Palmeiras player and current club director Cesar Sampaio, who attended Alves' funeral on Monday. "We have to take a stance to try to put an end to this right now."

There hadn't been a death linked to fan violence in Brazil since early last year, when a Corinthians supporter was killed after reportedly being ambushed by Palmeiras fans. Police believe Sunday's fight came in retaliation for that death, and Palmeiras supporters are already using social media networks to say they will avenge this weekend's killing.

Police were investigating reports that Sunday's fight was set up on the Internet. The Mancha Verde released a note saying the group was ambushed by the Corinthians supporters, but the Gavioes da Fiel denied the allegations.

The fans used iron bars, pieces of wood and rocks in the confrontation which lasted several minutes until riot police arrived to intervene. It happened several hours before the match between Corinthians and Palmeiras, several kilometers from the stadium.

There had been few incidents involving fan groups in the past few years, but several have been reported in recent months, including some between Corinthians and Palmeiras fans.

About a week ago, a 28-year-old fan of small club Guarani died from head injuries after fighting with Ponte Preta supporters in the city of Campinas, about 100 kilometers (60 miles) from Sao Paulo.

Federal budget cuts wiped out the $49 million Mentoring Children of Prisoners (MCP) program last September, effectively ending one of the signature domestic social service programs created under the George W. Bush administration's faith-based initiative.

But it's not the end of the Bush legacy of government partnerships with religious social service providers. Former President Bush's effort to make the social safety net more religious was one of the flash points of the culture wars that raged as he came to office in 2001: Funding religious groups well placed in communities to serve the needy was criticized for the potential for proselytizing and hiring bias using tax dollars.

Today, experts say the initiatives sparked more political debate than meaningful change to the social service landscape. Perhaps most important, the initiatives ? carried forward in different ways by the Obama administration ? "overcame the 'culture of resistance' " to such government-religious partnerships, noted a 2009 study of the program by the Nelson A. Rockefeller Institute of Government and the Roundtable on Religion and Social Welfare Policy.

"Of all the signature initiatives of the Bush era ... faith-based initiatives [policy] has managed to survive," says Lew Daly, a senior fellow at Demos, a New York policy research group. "That means Bush was onto something about this question of church-state partnerships."

Though a far cry from what Mr. Bush envisioned, what remains is a strong government infrastructure for mobilizing churches.

Faith-based initiatives under Bush were "about the symbolic politics of it all, and riling up [the GOP] base," says Rebecca Sager, a Loyola Marymount University sociologist and author of "Faith, Politics, & Power: The Politics of Faith-Based Initiatives." "It was showing you supported religious groups ... and all these wonderful things were going to happen without ... the real financial support to make that happen."

Under Bush, the White House Office of Faith-Based and Community Initiatives oversaw government-wide efforts to make it easier for religious groups ? particularly small community-based groups ? to compete for funding to deliver social services. In areas from feeding programs to treating drug addictions, rules were modified to help religious groups qualify for funding and maintain their religious character once funded.

Controversy swirled from the start. Activists for separation of church and state worried that government funds would be used for religious activities and protested a policy allowing faith-based groups to use religious considerations in hiring while receiving government funding.

Yet for all the hand-wringing and public debate, little changed in terms of how people receive social services. Organizations such as Catholic Charities, the Salvation Army, and Lutheran Social Services continue to receive substantial government funding, just as they have for decades.

Scientists describe humanity's global impact as 'The Great Acceleration' and offer ominous outlook: An uncertain future on a much hotter world

Time is running out to minimize the risk of setting in motion irreversible and long-term climate change and other dramatic changes to Earth's life support system, according to scientists speaking at the Planet Under Pressure conference, which began in London today.

The unequivocal warning is delivered on the first day of the four-day conference opening with the latest readings of Earth's vital signs.

In subsequent days at the meeting, nearly 3,000 experts spanning the spectrum of interconnected scientific interests, will examine solutions, hurdles and ways to break down the barriers to progress. The conference is the largest gathering of experts in development and global environmental changes in advance of June's UN "Rio+20" summit in Brazil.

"The last 50 years have without doubt seen one of the most rapid transformations of the human relationship with the natural world," says speaker Will Steffen, a global change expert from the Australian National University.

"Many human activities reached take-off points sometime in the 20th Century and sharply accelerated towards the end of the century. We saw a 'Great Acceleration'."

"It is the scale and speed of the Great Acceleration that is truly remarkable. This has largely happened within one human lifetime."

Key indicators of the planet's state, according to the speakers: higher levels of carbon dioxide in the atmosphere, phosphorus extraction and fertilizer production causing many large dead zones in coastal areas; rising air and ocean temperatures; melting sea ice, polar ice sheets and Arctic permafrost; rising sea levels and ocean acidification; biodiversity loss; land use changes; and growing consumption of freshwater supplies and energy by a growing global population, of which billions of people still lack even the most basic elements of well-being.

At a planetary level, humanity is altering the global carbon cycle, water cycle and nitrogen cycle, says Professor Steffen. Indeed, humans now produce more reactive nitrogen artificially than all natural processes on land.

"Where on Earth are we going?" he asks, underlining several potentially dangerous environmental "tipping points" foreseen, among them the melting of the polar ice sheets and the thawing of perennially frozen northern permafrost soils.

Current research estimates the permafrost alone stores the equivalent of roughly twice the carbon in the atmosphere, he says. Under a "high warming scenario," projected releases of greenhouse gas emissions from melting Arctic permafrost are the equivalent of 30-63 gigatonnes (Gt) of carbon dioxide by 2040, 232-380 Gt by 2100, and 549-865 Gt by 2300. By comparison, fossil fuel emissions today are roughly 10 Gt per year.

"The key point is: Either we turn around a lot of these trends the carbon dioxide trend, deforestation and so on or we allow them to continue and push beyond critical thresholds."

"There are signs that some drivers of global change are slowing or changing," says fellow speaker Professor Diana Liverman, co-Director of the Institute of the Environment at the University of Arizona and visiting Oxford University academic.

"Population growth is slowing and will level off; the intensity of energy and carbon required for a unit of production is declining; agricultural intensification is slowing and forests are starting to expand in some regions."

"On the other hand, average resource consumption per person, already high in some regions, is growing steeply in emerging economies even as many poor people cannot meet basic human needs.

"In some countries people are consuming far too much, including carbon, water and other resources embodied in trade. We have a long way to go to turn things around."

Says conference co-chair and UNESCO director of the science-policy division, Dr Lidia Brito: "If you like, our presenters today are akin to doctors saying 'look, you may not feel too sick at the moment but you've got high blood pressure, your cholesterol is going up, and your lifestyle is not conducive to good health.'"

"There is time to turn these trends around and promising, more positive messages will be delivered by colleagues in days to come. We look forward to discussions of our most promising options, the barriers to change and to a prescription for the future."

Scientists describe humanity's global impact as 'The Great Acceleration' and offer ominous outlook: An uncertain future on a much hotter world

Time is running out to minimize the risk of setting in motion irreversible and long-term climate change and other dramatic changes to Earth's life support system, according to scientists speaking at the Planet Under Pressure conference, which began in London today.

The unequivocal warning is delivered on the first day of the four-day conference opening with the latest readings of Earth's vital signs.

In subsequent days at the meeting, nearly 3,000 experts spanning the spectrum of interconnected scientific interests, will examine solutions, hurdles and ways to break down the barriers to progress. The conference is the largest gathering of experts in development and global environmental changes in advance of June's UN "Rio+20" summit in Brazil.

"The last 50 years have without doubt seen one of the most rapid transformations of the human relationship with the natural world," says speaker Will Steffen, a global change expert from the Australian National University.

"Many human activities reached take-off points sometime in the 20th Century and sharply accelerated towards the end of the century. We saw a 'Great Acceleration'."

"It is the scale and speed of the Great Acceleration that is truly remarkable. This has largely happened within one human lifetime."

Key indicators of the planet's state, according to the speakers: higher levels of carbon dioxide in the atmosphere, phosphorus extraction and fertilizer production causing many large dead zones in coastal areas; rising air and ocean temperatures; melting sea ice, polar ice sheets and Arctic permafrost; rising sea levels and ocean acidification; biodiversity loss; land use changes; and growing consumption of freshwater supplies and energy by a growing global population, of which billions of people still lack even the most basic elements of well-being.

At a planetary level, humanity is altering the global carbon cycle, water cycle and nitrogen cycle, says Professor Steffen. Indeed, humans now produce more reactive nitrogen artificially than all natural processes on land.

"Where on Earth are we going?" he asks, underlining several potentially dangerous environmental "tipping points" foreseen, among them the melting of the polar ice sheets and the thawing of perennially frozen northern permafrost soils.

Current research estimates the permafrost alone stores the equivalent of roughly twice the carbon in the atmosphere, he says. Under a "high warming scenario," projected releases of greenhouse gas emissions from melting Arctic permafrost are the equivalent of 30-63 gigatonnes (Gt) of carbon dioxide by 2040, 232-380 Gt by 2100, and 549-865 Gt by 2300. By comparison, fossil fuel emissions today are roughly 10 Gt per year.

"The key point is: Either we turn around a lot of these trends the carbon dioxide trend, deforestation and so on or we allow them to continue and push beyond critical thresholds."

"There are signs that some drivers of global change are slowing or changing," says fellow speaker Professor Diana Liverman, co-Director of the Institute of the Environment at the University of Arizona and visiting Oxford University academic.

"Population growth is slowing and will level off; the intensity of energy and carbon required for a unit of production is declining; agricultural intensification is slowing and forests are starting to expand in some regions."

"On the other hand, average resource consumption per person, already high in some regions, is growing steeply in emerging economies even as many poor people cannot meet basic human needs.

"In some countries people are consuming far too much, including carbon, water and other resources embodied in trade. We have a long way to go to turn things around."
